An Investigation of the Osteoclasts and their Precursors by Histological and Enzyme Histochemical Methods

Abstract
To investigate the osteoclasts and their precursors, the authors induced bone remodeling along the periosteum of right limb of the mandible by extracting the right upper molars in rats. At 1,2,3,4, and 5 days after induction of bone remodeling,
the
right mandibles were dissected and studied histologically and enzyme histochemically with nonspcific esterase(NSE), acid phosphatase(ACP), and alkaline phosphatase(ALP).
@ES The results were as follows:
@EN 1. In control group, periosteal surface was covered with osteoid tissues and osteblasts, and the osteoblasts showed positive reaction for the ALP. No osteoclasts could be seen.
2. In experimental group, osteoid tissues and the number of osteobalsts tended to decreased and disappeared completely after 3 day of induction. The mononuclear cell, however, were present at 1 day, and increased in number until 2 days after
induction.
They showed positive reaction for either NSE or ACP. From 3 days of induction, the NSE positive cells showed a gradual decrease while the ACP positive ones remained increase.
3. The osteoclasts were first observed at 2 days of induction and increased in number at 4 and 5days. These cells were strong positive for ACP, but weak to moderate positive or negative reaction for NSF.
4. Above findings may indicate that the resorption phase by the osteoclasts precedes the formation phase by the osteoblasts in bone remodeling, and progenitor cell of the osteoclasts is the mononuclear cell which belongs to the mononuclear
phagocyte
system.
